V Raziskovalni Skupini Drago\u0161 preu\u010dujemo odnos med temperentnimi fagi in bakterijami.&nbsp;<\/p>\n\n\n\n<p>Fagi lahko spremenijo fiziologijo, socialne interakcije in evolucijo svojega bakterijskega gostitelja. Ta odnos raziskujemo z razli\u010dnimi metodami, ki vklju\u010dujejo evolucijske, genetske in ekolo\u0161ke vidike interakcij, pri \u010demer uporabljamo bioinformatiko, molekularno biologijo in mikrobiolo\u0161ke pristope.&nbsp;<\/p>\n\n\n\n<p><strong>Na\u0161 model sistema fag-gostitelj <\/strong>je fag SPbetavirus in bakterija <em>Bacillus subtilis<\/em>, kozmpolitska bakterija, ki tvori spore in je znana po svojih lastnostih PGPR (pospe\u0161evalec rasti rastlin) ter probioti\u010dnih u\u010dinkih. <em>B. subtilis<\/em> je pomemben modelni organizem v raziskavah bakterij in se \u0161iroko uporablja v industriji za proizvodnjo encimov, antibiotikov in probiotikov. Raziskujemo ekstremne primere, kjer temperentni fagi spremenijo celi\u010dno steno gostitelja in preoblikujejo njegovo obliko.&nbsp;<\/p>\n\n\n\n<p><strong>Evolucija fagov prek rekombinacije<\/strong>&nbsp;<br>SPbetavirusi vsebujejo dolge homologne regije in se lahko med so\u010dasno oku\u017ebo rekombinirajo ter tvorijo hibridne variante. Prou\u010dujemo genomiko teh variant, morfologijo plakov in njihovo stabilnost, da bi bolje razumeli evolucijo znotraj fagnega rodu.&nbsp;<\/p>\n\n\n\n<p><strong>Raznolikost <em>Bacillus<\/em> profagov<\/strong>&nbsp;<br>Raziskujemo raznolikost vseh profagnih elementov, identificiranih v zaklju\u010denih genomih <em>B. subtilis<\/em>, ki so na voljo na NCBI, pa tudi v genomih iz lokalne zbirke na\u0161ega oddelka. Te elemente zdru\u017eujemo v funkcionalne taksonomske skupine in analiziramo njihovo raz\u0161irjenost ter raznolikost.&nbsp;<\/p>\n\n\n\n<p><strong>Nadzor gostitelja prek mehanizma regulatornega stikala<\/strong>&nbsp;<br>SPbetavirusi lahko delujejo kot regulatorno stikalo fagov, tako, da se integrirajo v funkcionalne bakterijske lokuse in jih ob tem inaktivirajo. Ob izrezu iz svojih vstavitvenih mest omogo\u010dijo ponovno vzpostavitev in izra\u017eanje prizadetih bakterijskih genov. Tak\u0161na integracija in izrez fagov vplivata na bakterijske fenotipe, kot so lastnosti bakterijskih spor. Razvili smo molekularna orodja za sledenje integracije in izreza fagov znotraj gostiteljskih celic. Raziskujemo dinamiko regulatornega izreza fagov pri razli\u010dnih bakterijskih sevih ter preu\u010dujemo fenotipske lastnosti, ki jih nadzorujejo regulatorni mehanizmi, na primer lastnosti spor.&nbsp;<\/p>\n\n\n\n<p><strong>Fagi kot nosilci bakteriocinov in posredniki medbakterijske tekmovalnosti<\/strong>&nbsp;<br>Na\u0161e delo je pokazalo, da fagi lahko nosijo biosintetske genske gru\u010de, ki kodirajo protimikrobne spojine, zaradi \u010desar igrajo aktivno vlogo v bakterijskem tekmovanju. Raziskujemo vlogo fagno kodiranih bakteriocinov pri vertikalnem in horizontalnem prenosu fagov ter preu\u010dujemo, kako lizogenija dolo\u010denih SPbetavirusov vpliva na sorodstveno diskriminacijo gostitelja.&nbsp;<\/p>\n\n\n\n<p><strong>Fagna in gostiteljska regulacija dinamike lizogenija-liza ter izra\u017eanja genov<\/strong>&nbsp;<br>Primerjamo dinamiko lizogenija-liza v zbirki lizogenov, ki se razlikujejo bodisi po fagnem, bodisi po gostiteljskem genetskem ozadju. Izvajamo tudi transkriptomske \u0161tudije za bolj\u0161e razumevanje molekularnih interakcij med fagi in gostiteljem, pri \u010demer se posebej osredoto\u010damo na gene, ki so pomembni v fazi lizogenije, ter na eksperimentalne pogoje, pri katerih profagi ka\u017eejo zanimive izra\u017ealne profile genov.&nbsp;<\/p>\n\n\n\n<p><strong>Sintenija genomov fagov in ohranjenost genov<\/strong>&nbsp;<br>Razvili smo <strong>Synphage<\/strong>, bioinformatsko orodje za vizualizacijo sintenije genov znotraj fagnega genoma.
